Description
Trex1 KO cell line is a spontaneously transformed mouse embryonic fibroblast (MEF) cell line. The line was derived from Trex1-/- knockout mouse by repeated passaging in culture using standard techniques for culturing primary MEFs. Trex1 is the major 3' DNA exonuclease in mammalian cells, and mutations in the human TREX1 gene can cause Aicardi-Goutières syndrome, characterized by perturbed immunity. Trex1-deficient cells show defective G1/S transition, with increased cell doubling time and chronic ATM-dependent checkpoint activation, even in the absence of exogenous stress, correlating with persistent single-stranded DNA molecules produced in S phase, which accumulate in the ER. Trex1 KO can be used to study novel interconnections between DNA replication, DNA damage checkpoint signalling and antiviral-like autoimmune responses.
