Description
The cell line TRA-171 has been established from freshly hatched larvae of Toxorhynchites amboinensis. After eggs had been surface-sterilised, they were allowed to hatch in MM / VP12 medium and were mechanically damaged at the anterior or posterior ends of the body in 0.25% trypsin. After washing and cultivation at 22 - 25°C small patches of cells were visible leading to the isolation of the cell line TRA-171 after 72 days in culture. The cells have been adapted to grow in a 11 mixture of Leibovitz L-15 and Mitsuhashi and Maramorosch media plus supplements. Four serotypes of laboratory adapted and three type strains (1, 2 and 3) of un-adapted dengue virus will replicate in the cells at 28°C.
