Description
Two different continuous cell lines were established from a plaque biopsy of an 82-year old male with mycosis fungoides stage II by inclusion of IL-2 and IL-4 in the culture medium My-La, CD4+, (referred to in the literature as My-La, marker chromosome) and My-La, CD8+ (referred to in the literature as My-La 46,XY-18+i(18q) (ECACC catalogue no. 95051033). My-La CD4+ is strictly dependent on exogenous IL-2 and IL-4 in the culture medium whereas My-La CD8+ on IL-2 only. Both show characteristic chromosome markers and are double homozygous for HLA-A1.B8. My-La CD8+ expresses beta-13 of the T cell receptor and phenotypically weak staining was observed for CD2, CD3, CD8, CD25, CD71 and TCR-1 alpha / ß. My-La CD4+ expresses beta-18 of the T cell receptor plus staining for CD2, CD3, CD4,CD25, CD71, HLA-DR and TCR-1 alpha/ ß. There has been no evidence of HTLV-I infection in the cell lines and serum from the patient did not demonstrate antibodies against HTLV-I and HTLV-II. Both cell lines show a low viability after resuscitation from frozen and a long lag time phase until exponential growth is reached.
Quality Control
Cell lines were tested and found to be free of mycoplasma, bacterial, viruses, and other toxins. All cells were above 95% viability before freezing.
